India ke ek aam hafte ke teen scenes socho. Maharashtra ka ek cotton farmer apne khet mein apne pita ke muqable bahut kam spray karta hai. Type 1 diabetes wali ek school ki ladki lunch se pehle insulin inject karti hai. Ek laboratory ek aise sample mein viral infection report karti hai jo patient ko zara sa bhi fever hone se pehle liya gaya tha. Har scene chupchaap biotechnology par tika hai.
Industrial sense mein biotechnology (जैव प्रौद्योगिकी) ka matlab hai living cells, ya unse nikale gaye molecules, ka use karke bade scale par useful products banana. Microbes, plants aur animals, jinmein se kai engineered hain, aaj polluted soil aur sewage saaf karne, biofuels banane, food aur detergents ke liye enzymes banane, aur vaccines, medicines, test kits aur behtar crops dene mein madad karte hain.
Doosra, woh conditions engineer karo jinmein woh catalyst sabse tez kaam kare; yeh bioreactor ka kaam hai. Teesra, product ko recover aur purify karo jab tak woh bechne, khaane ya inject karne layak saaf na ho jaaye; yeh hai downstream processing.
3/3Downstream processing product ko alag, purify aur pack karta hai.
Teen panels left se right chalte hain. Pehle step mein sirf pehla panel dikhta hai: ek rod-shaped bacterium jismein inserted gene wala plasmid hai, aur ek enzyme, label hai 1 Catalyst, best microbe. Doosre step mein ek arrow broth mein bubbles aur microbes wale stirred tank tak jaata hai, label hai 2 Conditions, bioreactor. Teesre step mein doosra arrow ek purification column tak jaata hai jiske coloured bands ek capped vial mein tapakte hain, label hai 3 Downstream, purify and pack.
Yeh chapter in tools ke peeche-peeche laboratory se bahar jaata hai: kheton mein (Section 10.1), hospitals aur clinics mein (Section 10.2), research aur medicine ke liye banaye gaye animals mein (Section 10.3), aur aakhir mein un debates mein ki living cheezon ko kaun badal sakta hai aur unka maalik kaun ho sakta hai (Section 10.4).
10.1 Agriculture mein Biotechnology ke Applications
India ko 1.4 billion se zyada logon ka pet lagbhag utni hi kheti ki zameen se bharna hai jitni woh 1970s mein jotta tha. Mote taur par us zameen se zyada food nikaalne ke teen tareeke hain, aur har ek ki apni keemat hai.
Zyada food ke teen raaste
Agrochemical farming
- Synthetic fertilisers aur pesticides yield badhate hain.
- Chhote farmers ke liye mehenga; zyada use soil, paani aur useful insects ko nuksaan pahunchata hai.
Organic farming
- Manure, compost, crop rotation aur biological pest control.
- Soil life ke liye narm, lekin yield aksar kam aur mehnat zyada.
GM-crop farming
- Crops mein aise genes hote hain jo unhe bachate ya behtar banate hain.
- Pesticide use kam kar sakta hai; safety, seed cost aur regulation ke sawaal uthata hai.
Lekin ab yeh badhat dheemi pad gayi hai. Agrochemicals itne mehenge hain ki kai chhote farmers afford nahi kar paate, zyada use karne par woh soil aur paani ko nuksaan pahunchate hain, aur conventional breeding dheemi hai aur sirf wahi traits add kar sakti hai jo un plants mein hain jinhe crop ke saath cross kiya ja sake. Biotechnology ne aur raaste khole, jinki shuruaat tissue ke chhote tukdon se poore plants ugaane se hui.
Tissue culture aur micropropagation
Ek leaf se ek disc kaato, use sterilise karo, ek band flask mein nutrients ki jelly par rakho, aur sahi hormones ke saath woh roots, shoots aur flowers wala naya plant ban sakta hai. Shuruaati tukde ko explant kehte hain aur is technique ko tissue culture (ऊतक संवर्धन).
ExampleCarrot root ka ek cell cluster leaves, flowers aur seeds wale carrot plant mein badalta hai.
Culture medium gel mein jama hua ek complete diet hai. Band vessel mein carbon dioxide kam aati hai, isliye explant photosynthesis se kaafi food nahi bana paata aur use ek carbon source chahiye, aam taur par sucrose. Inorganic salts nitrogen, phosphorus, potassium aur trace elements dete hain; vitamins aur amino acids metabolism ko support karte hain; agar mixture ko jelly mein jama deta hai. Asli faisla karne wale ingredients growth regulators hain, ek auxin aur ek cytokinin. Inka ratio development ko disha deta hai: balance hone par aksar callus banta hai (dividing cells ka ek unorganised lump), zyada cytokinin shoots ko badhava deta hai aur zyada auxin roots ko.
5/5Baar-baar subculture se hazaaron identical plants milte hain, somaclones, jinhe harden karke gamlon mein lagaya jaata hai.
Pehle step mein ek pattedar parent plant ke ek chhote tukde par dashed box bana hai, aur ek arrow ek capped culture tube tak jaata hai jismein woh tukda, explant, yellow gel par rakha hai. Doosre step mein ek panel aata hai jo sterile medium ki list deta hai: sucrose aur mineral salts, vitamins aur amino acids, auxin aur cytokinin. Teesre step mein doosri tube mein ek bedhab lump dikhta hai, callus. Chauthe step mein teesri tube mein leaves aur roots wala ek chhota plantlet hai. Aakhri step mein ek label par thousands in months likha hai aur paanch identical potted plants ki line aati hai, jinka label somaclones hai.
Agar bante hue shoots ko kaat kar baar-baar fresh medium mein le jaaya jaaye, to ek hi explant kuch mahino mein hazaaron plantlets de sakta hai. Yeh hai micropropagation (सूक्ष्मप्रवर्धन). Kyunki har plantlet parent se sirf mitosis se banta hai, sab mein parent jaise hi genes hote hain; aise plants ko somaclones kehte hain. Isliye ek grower ek shaandaar plant ko bina genes ki us uthal-puthal ke multiply kar sakta hai jo seeds laate hain. (Kabhi-kabhi culture process khud chhote genetic changes kar deta hai, jise somaclonal variation kehte hain, isliye growers apne stocks check karte hain.)
India mein banana iska ek zabardast example hai: Grand Naine variety ke tissue-culture plantlets bade scale par lagaye jaate hain kyunki woh uniform, healthy hote hain aur ek saath harvest ke liye taiyaar hote hain. Tomato, apple, potato, sugarcane, orchids aur kai forest trees bhi commercially micropropagate kiye jaate hain.
Meristems se virus-free plants
Cuttings, tubers ya suckers se ugaayi gayi crops mein viruses ek ziddi problem hain, kyunki har naye plant ko parent ka infection virasat mein milta hai. Phir bhi buri tarah infected plant mein bhi shoot tip par dividing cells ka chhota sa dome (apical meristem) aur leaf axils ke buds (axillary meristems) aam taur par virus se free hote hain. Viruses zyadatar plant ke vascular tissue se hokar chalte hain, jo meristem ke andar abhi bana nahi hota, aur meristem ke cells itni tezi se divide hote hain ki virus unmein phail nahi paata.
Isse growers ko bachne ka raasta milta hai (Fig. 10.3). Microscope ke neeche ek technician millimetre ke ek chhote se hisse jitna lamba meristem nikaalta hai aur use culture medium par ugaata hai; jo plant banta hai woh shuru se saaf hota hai. Potato seed-tuber programmes, sugarcane nurseries aur banana producers sab meristem culture use karte hain, aur phir healthy plants ko multiply karne ke liye micropropagation.
3/3Medium par ugaane se meristem ek virus-free plant ban jaata hai.
Pehle step mein left mein ek plant khada hai jiski neeche ki leaves peeli hain aur uske stem aur leaves par chhote hexagonal virus particles bikhre hain, lekin gol ghere wali shoot tip mein ek bhi nahi; labels par virus everywhere except the tip likha hai. Doosre step mein ek magnified circle dome-shaped meristem dikhata hai jiske aar-paar dashed cut line hai, label hai meristem, cut about 0.3 mm. Teesre step mein ek arrow ek culture tube tak jaata hai jismein ek healthy hara plantlet ugta hai, label hai virus-free new plant.
Somatic hybridisation
Breeders aam taur par do plants ko cross-pollination se jodte hain, jo sirf kareebi relatives ke beech kaam karta hai. Tissue culture is rukawat se bachne ka raasta deta hai. Cellulase aur pectinase jaise enzymes cell walls ko digest kar dete hain, aur peeche naked protoplasts (जीवद्रव्यक) bachte hain jo sirf apni plasma membranes se ghire hote hain. Beech mein walls na hone se do alag plants ke protoplasts ko fuse karaya ja sakta hai, polyethylene glycol (PEG) jaise chemical ya ek chhote electric pulse se.
Fused cell mein dono parents ka genetic material hota hai. Culture mein woh apni wall dobara banata hai, divide hota hai aur ek plant mein regenerate ho sakta hai jise somatic hybrid kehte hain; is process ko somatic hybridisation kehte hain (Fig. 10.4). Sabse mashhoor koshish mein tomato aur potato ke protoplasts fuse karke pomato banaya gaya. Woh uga, lekin usne kabhi tomatoes ki acchi fasal aur potatoes ki acchi fasal ek saath nahi di, isliye uska koi commercial use nahi hua. Somatic hybrids wild relatives se potato jaisi crops mein disease resistance laane mein zyada successful rahe hain.
3/4PEG ya electric pulse unhe fuse karke ek hybrid cell banata hai.
Pehle step mein do square plant cells aamne-saamne hain, har ek ki moti hari wall hai: solid outline wala tomato cell aur dashed outline wala potato cell. Doosre step mein walls gayab ho jaati hain, do gol protoplasts bachte hain, aur label kehta hai enzymes digest walls. Teesre step mein protoplasts khisak kar paas aate hain aur unki jagah dono nuclei wala ek bada oval hybrid cell aa jaata hai, label hai fused by PEG or a pulse. Aakhri step mein ek arrow ek plant tak jaata hai jiske soil line ke upar gol fruits aur neeche tubers hain, label hai pomato, a somatic hybrid plant.

Genetically modified crops
Kya genes ka hamara gyaan farmers ko kam chemicals use karke zyada fasal lene mein madad kar sakta hai? Tissue culture poore genomes ko copy ya combine karta hai; genetic engineering zyada precise hai aur sirf ek ya kuch chune hue genes ko move karti hai. Jin plants, animals, bacteria ya fungi ke genes is tarah badle gaye hain, woh genetically modified organisms hain (GMOs, आनुवंशिकतः रूपांतरित जीव).
Aisa organism jiska genetic material mating ya natural recombination ki jagah genetic engineering se badla gaya hai, jaise kisi doosri species ka gene add karke.
ExampleBt cotton, jismein ek soil bacterium ka insecticidal gene hota hai.
GM crops aisi problems solve karne ke liye design kiye gaye hain jinse normal breeding jujhti rahi:
- Built-in pest protection, taaki farmers kam insecticide spray karein.
- Abiotic stress mein survival: drought, thand, garmi ya namkeen soil.
- Behtar nutrition: golden rice β-carotene banata hai, jise hamara body vitamin A mein badal deta hai.
- Fertiliser ka kifayati use: jo plants nitrogen ya phosphorus zyada efficiently lete hain, woh soil ko kam khaali karte hain.
- Lambi shelf life: jo fruit dheere naram hone ke liye engineer kiya gaya hai, woh transport aur storage jhel leta hai, isliye kam food barbaad hota hai.
- Factories ke roop mein plants: aisi crops jo industrial starches, biofuels, ya vaccines aur doosri drugs dene ke liye engineer ki gayi hain.
Do applications dikhate hain ki aisi crops kaise banti hain: plants jo apna insecticide khud banate hain, aur plants jo ek parasite ke genes ko silence kar dete hain.
Bt cotton
Bacillus thuringiensis (short mein Bt) ek soil bacterium hai. Jab yeh spores banata hai, to kuch proteins ko microscopic crystals mein bhi pack karta hai jo kuch insects ke liye zehreele hote hain. Farmers, organic farmers bhi, dashakon se Bt ko biological pesticide ki tarah spray karte aaye hain. Genetic engineering ek kadam aage gayi: bacterium se ek toxin gene isolate karke crop plants mein insert kiya gaya, jo ab apni leaves, stems aur bolls mein khud insecticidal protein banate hain. Bt cotton, Bt maize (corn) aur Bt soybean bade paimane par ugaaye jaate hain; rice, potato, tomato aur brinjal ke Bt versions bhi banaye gaye hain, aur Bangladesh ke farmers Bt brinjal ugaate hain.
Har strain ka toxin insects ke ek chhote group par kaam karta hai. Cotton mein target moths ke caterpillars hain, yaani lepidopterans (bollworms, budworms, armyworms). Doosre Bt strains beetle larvae (coleopterans) ya flies aur mosquitoes ke larvae (dipterans) ke liye jaanleva hain. Zyadatar doosre animals, insaan bhi, par iska asar nahi hota.
Jawaab yeh hai ki toxin ek inactive protoxin ke roop mein store hota hai. Bt leaf chabaane wala caterpillar crystals nigal leta hai. Uska midgut bahut alkaline hota hai (kai caterpillars mein lagbhag pH 10), jo crystals ko ghol deta hai, aur uske apne gut enzymes protoxin ko kaat kar active toxin bana dete hain. Active toxin midgut ki lining wale epithelial cells par specific receptor proteins se bind hota hai aur unki membranes mein ghuskar pores bana deta hai. Paani aur ions tezi se andar aate hain, cells phool kar phat jaate hain (lyse), gut wall toot jaati hai, aur insect khaana band kar deta hai aur kuch dino mein mar jaata hai (Fig. 10.5). Hamara stomach acidic hai aur hamare gut cells mein woh receptors nahi hote, isliye protein bas digest ho jaata hai.
3/4Toxin midgut cells ke receptors se bind hota hai aur unki membranes mein pores banata hai.
Drawing ka upar wala hissa gut space hai, label hai alkaline, pH about 10; uske neeche paanch lambe gut-lining cells ki line hai jinmein nuclei hain aur upar chhote projections hain. Pehle step mein gut space mein teen diamond-shaped crystals tair rahe hain, label hai protoxin crystals eaten with the leaf. Doosre step mein crystals gayab hain aur unki jagah bahut saare chhote active toxin molecules bikhre hain, label hai crystals dissolve and enzymes trim them. Teesre step mein har cell ke upar ek toxin molecule baitha hai aur paani ka ek arrow andar behta hai, label hai binds the gut lining and punches pores. Aakhri step mein cells phoole hue hain aur unpar daraar ke nishaan hain, label hai cells swell and burst, gut fails and the insect dies.
Bt toxins ko Cry proteins kehte hain (crystal se), aur unhe encode karne wale genes cry genes hain. Saikdon jaane jaate hain, har ek insects ke ek khaas group ke against active, isliye engineers woh gene chunte hain jo crop aur uske main pest se match kare (Table 10.1). Tumhari textbook naam roman numerals mein likhti hai, jaise cryIAc; aaj ki naming arabic numerals use karti hai, cry1Ac. Dono ka matlab same gene hai.
| Gene | Main target | Crop |
|---|---|---|
| cryIAc (cry1Ac) | bollworms | cotton |
| cryIIAb (cry2Ab) | bollworms | cotton |
| cryIAb (cry1Ab) | corn borer | maize |
India ne cry1Ac wale apne pehle Bt cotton hybrids 2002 mein approve kiye. Do genes, cry1Ac aur cry2Ab, wali second generation 2006 mein aayi, taaki pest ko ek saath do toxins ka resistance develop karna pade. Aaj India ke cotton area ke nau-dasve hisse se zyada mein Bt hybrids boye jaate hain, aur Bt cotton abhi bhi desh mein commercially ugaayi jaane wali akeli GM crop hai.
Bt crops ki ek kamzori hai: evolution. Pink bollworm, jo cotton boll ke andar chhup kar khaata hai, India ke bade hisse mein dono toxins ke liye resistant ho gaya hai, aur farmers ko uske against phir se spray karna pada hai. Resistance sabse tezi se tab phailta hai jab district ke har plant mein toxin ho, kyunki tab sirf resistant moths hi bach kar mate karte hain. Isliye growers se non-Bt cotton ka ek refuge rakhne ko kaha jaata hai, jo aaj aksar seed packet mein mile non-Bt seeds ke roop mein aata hai, taaki susceptible moths bachein aur rare resistant moths ke saath mate karein (Fig. 10.6).

Pest-resistant plants: RNA interference
Har pest insect nahi hota. Nematodes naam ke chhote roundworms soil ke neeche se crops par hamla karte hain. Sabse nuksaandayak mein se ek hai root-knot nematode, Meloidogyne incognita. Iske larvae tobacco aur kai doosri crops ki roots mein ghus kar root cells ko feeding sites bana dete hain; roots phool kar knots (galls) ban jaati hain, paani aur minerals shoots tak pahunchna band ho jaate hain, aur harvest ka bada hissa barbaad ho jaata hai.
Is baar hathiyaar khud eukaryotic cells se udhaar liya gaya. RNA interference (RNAi) mein ek double-stranded RNA (dsRNA) har us gene ko switch off kar deta hai jiske messenger RNA ka sequence usse match karta hai. Nature mein aisa dsRNA kahan se aata hai? Kai viruses ka genetic material RNA hota hai aur multiply hote waqt woh dsRNA banate hain, aur kuch transposons (‘jumping genes’) RNA ke zariye apni copy banate hain. Healthy cells mein lamba dsRNA kam hi hota hai, isliye yeh danger signal ka kaam karta hai, aur RNAi cell ko matching messages nasht karne deta hai.
Eukaryotic cells mein ek specific gene ko us gene ke mRNA se match karne wale double-stranded RNA dwara silence karna; mRNA nasht ya block ho jaata hai, isliye koi protein nahi banta.
ExampleTobacco plants jo nematode gene se match karne wala dsRNA banate hain, jisse khaane wale nematode ka apna gene silence ho jaata hai.
Cell ke andar Dicer naam ka enzyme dsRNA ko lagbhag 21–23 nucleotides lambe chhote tukdon mein kaat deta hai, jinhe small interfering RNAs (siRNAs) kehte hain. RISC naam ka ek protein complex har tukde ka ek strand guide ki tarah rakhta hai, complementary sequence wala koi bhi mRNA dhoondhta hai, aur use kaat deta hai ya uska translation block kar deta hai. Gene abhi bhi maujood hai, lekin uska message kabhi protein nahi banta.
Plan yeh tha ki tobacco plant khud trigger banaye. Researchers ne aise genes chune jinke bina nematode zinda nahi reh sakta, aur Agrobacterium ko vector bana kar tobacco ko nematode sequence ki aisi copy di jise plant dono directions mein transcribe kare, jisse sense RNA aur complementary antisense RNA bane. Dono strands plant ke cells ke andar pair hokar dsRNA ban gaye (Fig. 10.7). Jab nematode ne roots khaayi to usne yeh dsRNA le liya, uski apni RNAi machinery ne matching mRNA ko silence kar diya, aur parasite zinda nahi reh saka. In transgenic plants ki roots un galls se kaafi had tak free rahin jo normal plants ki roots par chhaye the.
5/5Guide strand matching nematode mRNA dhoondhta hai, jo kat jaata hai: koi protein nahi, aur parasite mar jaata hai.
Do panels aamne-saamne hain: left mein plant root cell aur right mein nematode. Pehle step mein plant nucleus mein ek transgene hai, jisse ek solid sense RNA strand aur ek dashed antisense RNA strand nikalte hain. Doosre step mein dono strands paas aate hain aur rungs unhe ek ladder mein jod dete hain, label hai dsRNA, pairs form a double strand. Teesre step mein ek arrow dsRNA ko nematode panel mein le jaata hai, label hai taken in while feeding. Chauthe step mein lamba dsRNA teen chhote tukdon mein kat jaata hai, label hai Dicer cuts into siRNAs, aur ek lamba nematode mRNA dikhta hai. Aakhri step mein ek guide strand mRNA ke neeche hai, mRNA cross mark ke saath kata hai, aur labels par siRNA guide finds the match, mRNA cut, no protein made, nematode dies likha hai.

10.2 Medicine mein Biotechnology ke Applications
Recombinant DNA technology se pehle kai protein medicines animal organs ya donate kiye gaye human blood se nikaalni padti thin. Supply limited thi, batches alag-alag hote the, ek foreign protein immune system ko bhadka sakta tha, aur donor material mein chhupe infections ho sakte the. Engineered cells ne yeh badal diya. Woh kisi bhi size ke fermenters mein human proteins banate hain, jo hamare apne proteins jaise identical hain, aur kyunki body in proteins ko apna pehchanti hai, allergic aur immune reactions rare ho jaate hain aur koi donor infection andar nahi aa sakta.
Tumhari textbook duniya bhar mein approve hue lagbhag 30 recombinant therapeutics ki baat karti hai; aaj yeh ginti kai sau tak pahunch gayi hai. India inmein se kai banata hai, jinmein low-cost biosimilars bhi hain, aur 1997 mein Hyderabad ki ek company ne hepatitis B ke against India ki pehli desi recombinant vaccine launch ki. Kuch examples Table 10.2 mein list kiye gaye hain.
| Product | Kis liye use | Kahan banta hai |
|---|---|---|
| Human insulin | diabetes | E. coli ya yeast |
| Growth hormone | growth hormone ki kami | E. coli |
| Erythropoietin | kidney failure mein anaemia | hamster ovary cells |
| Hepatitis B vaccine | hepatitis B se bachaav | yeast |
| Tissue plasminogen activator | heart attack aur stroke mein clots gholna | hamster ovary cells |
10.2.1 Genetically Engineered Insulin
Insulin, pancreas ke beta cells ka ek hormone, body cells ko blood se glucose lene deta hai. Type 1 diabetes wale log lagbhag bilkul insulin nahi banate aur unhe roz insulin chahiye; lambe samay se type 2 diabetes wale kai logon ko bhi iski zaroorat padti hai. 2023 mein publish hui ek ICMR study ke andaaze se lagbhag 101 million Indians ko diabetes hai, isliye demand bahut badi hai.
1921 mein discovery ke baad lagbhag saath saal tak diabetes wale log abattoirs se ikattha kiye gaye pigs aur cattle ke pancreas glands se purify kiye gaye insulin par nirbhar the. Animal hormone isliye kaam karta hai kyunki woh lagbhag same molecule hai: pig insulin hamare insulin se 51 mein se ek amino acid par alag hai, aur cattle insulin teen par. Lekin kuch patients ne ise foreign protein maan kar react kiya, allergies ya antibodies ke saath, aur supply meat trade par depend karti thi. Human insulin banane wala bacterium dono problems solve kar deta: zyada insulin chahiye, to bas zyada bacteria ugaao.
Mature insulin ek chhota protein hai: 21 amino acids ki chain A aur 30 ki chain B, jo do disulphide bridges (–S–S–) se judi hain, aur chain A ke andar ek teesra bridge hai. Lekin pancreas do chains alag-alag nahi banata. Woh ek chain banata hai, proinsulin, ek pro-hormone jo processing ke baad hi active hota hai (pepsinogen se compare karo, jo tabhi active pepsin banta hai jab uska ek hissa kaat diya jaaye). Proinsulin mein ek connecting hissa, C peptide, chain B ke end ko chain A ki shuruaat se jodta hai. Chain fold hoti hai, bridges bante hain, aur phir enzymes C peptide ko kaat kar nikaal dete hain, jisse mature insulin release hota hai (Fig. 10.8).
2/3Chain fold hoti hai aur teen disulphide bridges bante hain.
Pehle step mein proinsulin teen hisson wali ek lambi horizontal bar ki tarah bana hai: 30 amino acids ki B chain, 31 ka C peptide aur 21 ki A chain. Doosre step mein chain fold hai: B chain upar hai, A chain uske neeche, aur C peptide right mein ek mota U-shaped link banata hai jo dono ko jodta hai; do yellow S–S bridges A aur B ko jodte hain, teesra chain A ke andar loop banata hai, aur ek label kehta hai 3 disulphide bridges. Aakhri step mein jahan C peptide chains se juda tha wahan cross marks aate hain, C peptide neeche right ki taraf khisak jaata hai jiska label free C peptide hai, aur bache hue pair ka label hai insulin, A plus B equals 51.
Proinsulin ke hisse gino
Human proinsulin 86 amino acids ki ek single chain hai. Mature insulin mein chain A (21 amino acids) aur chain B (30) hoti hai. (a) Proinsulin ke mature hone par kitne amino acids hataye jaate hain? (b) C peptide khud 31 amino acids lamba hai. Kitne aur hataye jaate hain, aur woh wahan kyun ho sakte hain? (c) Har disulphide bridge do cysteines ko jodta hai. Apne teen bridges ke liye insulin ko kitne cysteines chahiye?
Pehle khud try karo. Phir solution ek-ek step karke kholo.
Bacteria yeh processing nahi kar sakte, aur recombinant DNA technology se insulin banane mein yahi sabse badi rukawat thi. Iska hal, jo American company Eli Lilly ne Genentech mein develop kiye gaye methods se apnaya, yeh tha ki do chains alag-alag bacterial cultures mein banayi jaayein aur baad mein jodi jaayein. Har chain ke liye ek chhota synthetic gene banaya gaya, aur har gene apne alag E. coli plasmid mein gaya, isliye bacteria ke ek batch ne A chains banayi aur doosre ne B chains. Purification ke baad chains ko aisi conditions mein milaya gaya jinmein sahi disulphide bridges bante hain, aur result tha human insulin (Fig. 10.9). 1982 mein USA mein approve hokar aur 1983 se bazaar mein aakar, yeh recombinant DNA technology se bani pehli medicine thi.
4/4Milane par chains disulphide bonds se jud jaati hain: human insulin.
Figure mein do lanes hain, upar A aur neeche B. Pehle step mein har lane mein inserted gene wala ek plasmid ring hai, labels A gene aur B gene. Doosre step mein arrows do rod-shaped E. coli cells tak jaate hain, har ek mein uska plasmid aur kai chhote chain molecules hain: A ke liye solid, B ke liye dashed. Teesre step mein arrows do test tubes tak jaate hain, har ek mein purified chains hain, label hai pure. Aakhri step mein dono tubes ke arrows right mein milte hain, jahan ek lambi B chain aur ek chhoti A chain ek ke upar ek do yellow bridges se judi hain, label hai insulin, S–S bonds.
Product ka amino acid sequence bilkul natural hormone jaisa hai. Baad ke processes microbes mein khud proinsulin banate hain aur factory mein enzymes se C peptide kaat dete hain. Kai manufacturers, jinmein se kai India mein hain, ab E. coli ya yeast mein insulin banate hain, saath hi insulin analogues bhi jinmein ek ya do amino acids jaan-boojh kar badle jaate hain taaki insulin tez kaam kare ya zyada der tak chale.

10.2.2 Gene Therapy
Inherited diseases ki zyadatar medicines faulty gene ke effects ka ilaaj karti hain, jaise woh protein inject karke jo patient nahi bana sakta. Gene therapy (जीन चिकित्सा) jad par vaar karti hai: missing product baar-baar dene ki jagah, woh use banane ke instructions deti hai. Gene ki ek working copy patient ke cells mein pahunchayi jaati hai, jahan woh woh kaam karti hai jo faulty copy nahi kar paati.
Kisi disease, khaas kar inherited disease, ka ilaaj patient ke apne cells mein gene ki working copy pahunchakar karna, taaki woh faulty copy ki jagah le ya uski kami poori kare.
ExampleEk bachche ke lymphocytes ko working ADA gene dena.
Aam delivery vehicles disabled viruses hote hain. Viruses naturally apne genes ko un cells mein le jaate hain jinhe woh infect karte hain; vector mein disease paida karne wale genes hata diye jaate hain aur unki jagah therapeutic gene aa jaata hai.
Pehli approved clinical gene therapy 1990 mein USA mein hui. Patient, jo tab chaar saal ki thi, ko adenosine deaminase (ADA) deficiency thi. ADA ek enzyme hai jo deoxyadenosine ko saaf karta hai, jo DNA breakdown ka by-product hai. Iske bina by-product jama hota jaata hai aur developing lymphocytes ko maar deta hai, isliye immune system mushkil se kaam karta hai aur bachcha ek ke baad ek infection se beemar padta hai; yeh condition severe combined immunodeficiency (SCID) ka ek form hai. Yeh dono parents se mile faulty ya missing ADA gene ki wajah se hoti hai.
ADA deficiency wale bachchon ko working enzyme ke regular injections (enzyme replacement therapy) se zinda rakha ja sakta hai, lekin yeh sirf kuch had tak madad karte hain aur zindagi bhar chalne chahiye. Bone marrow transplant disease ko cure kar sakta hai, lekin sirf tab jab acchi tarah match karne wala donor mile, aur kai bachchon ke paas koi nahi hota.
1990 ki team ne body ke bahar kaam kiya (Fig. 10.10). Ladki ke blood se lymphocytes alag karke culture mein multiply kiye gaye; ek disabled retrovirus se unmein working ADA gene, cDNA ke roop mein, insert kiya gaya; aur engineered cells ko dheere-dheere wapas uske bloodstream mein daala gaya, jahan unhone ADA banaya. Lekin mature lymphocytes hamesha divide nahi hote rehte, isliye unki ginti ghat gayi aur har kuch mahine mein naye infusions ki zaroorat padti thi.
3/5Ek retroviral vector working ADA cDNA ko cells mein le jaata hai.
Left mein ek patient ki simple figure khadi hai, label hai patient, no ADA. Pehle step mein ek arrow blood cells ko ek culture dish tak le jaata hai jismein do gol lymphocytes hain, label hai lymphocytes grown outside. Doosre step mein multiply hote hue aur lymphocytes dish bhar dete hain. Teesre step mein right mein chhota gene liye ek hexagonal retrovirus aata hai, label hai retrovirus carries ADA gene, ek arrow dish mein jaata hai, aur lymphocytes ka rang badal jaata hai, unhe dashed outlines aur square nuclei milte hain jo dikhate hain ki unmein gene hai. Chauthe step mein ek arrow engineered cells ke infusion bag tak aur doosra arrow wapas patient tak jaata hai, label hai infused back. Aakhri step mein label par repeat, engineered cells die out likha hai.
Lambe samay ke cure ke liye gene un cells mein hona chahiye jo naye lymphocytes banate rehte hain. Tumhari textbook batati hai ki early embryonic stage par cells mein gene daalna permanent ho sakta hai. Lekin embryo ko badalne se woh eggs ya sperm bhi badal jaate hain jo woh baad mein banayega, isliye change aane wali generations tak jaayega. Aisi germline gene therapy India ki gene therapy ki national guidelines (ICMR, 2019) ke tahat allowed nahi hai aur zyadatar deshon mein banned ya sakhti se restricted hai. Asal mein jo raasta use hota hai woh hai bone marrow ke patient ke apne blood banane wale stem cells: ek baar theek karne par woh healthy lymphocytes banate rehte hain. ADA deficiency ke liye is tarah ka treatment 2016 mein Europe mein approve hua.

10.2.3 Molecular Diagnosis
Socho ek aisa virus jo fever laane mein do hafte leta hai. Un do hafton mein patient theek mehsoos karta hai, doosron ko infect kar sakta hai, aur use treatment se pehle hi fayda mil sakta tha. Jo tests disease ke effects dhoondhte hain (serum ya urine mein changes) ya jinhe bahut saare microbes chahiye (microscope slide, culture), woh aam taur par tabhi positive aate hain jab pathogen multiply ho chuka ho aur symptoms dikhne lagein. Molecular methods seedhe pathogen ke apne molecules, uske DNA, RNA ya proteins, dhoondhte hain, aur unhe tab bhi pakad sakte hain jab woh bahut kam hon. Teen tools sabse important hain: recombinant DNA probes, polymerase chain reaction (PCR) aur ELISA (Table 10.3).
| Method | Kya dhoondhta hai | Khoobi |
|---|---|---|
| PCR | Pathogen ka DNA ya RNA, ya ek mutated gene | target ko baar-baar copy karta hai |
| Labelled probe | ek khaas DNA sequence | sirf matching sequence se pair karta hai |
| ELISA | antigens, ya unke against antibodies | ek enzyme har binding ko colour mein badal deta hai |
PCR yeh problem copy karke solve karta hai. Har cycle target sequence ko zyada se zyada double kar sakta hai (RNA virus ko pehle reverse transcriptase DNA mein copy karta hai). Das cycles use lagbhag hazaar guna kar dete hain ( = 1024 hai), aur tees cycles lagbhag ek billion guna. Isliye mutthi bhar copies bhi itni ho jaati hain ki detector unhe dekh sake, symptoms aane se bahut pehle (Fig. 10.11).
PCR ka use infants mein HIV infection confirm karne aur patients mein virus ki maatra track karne, tuberculosis aur uske drug resistance ka pata lagane mein hota hai, aur COVID-19 pandemic ke dauraan yeh SARS-CoV-2 ka standard test tha. Tuberculosis ke liye India ke apne chip-based PCR test, Truenat, ko 2020 mein World Health Organization ne endorse kiya. PCR cancer cells mein aur sickle-cell anaemia aur thalassaemia jaise inherited disorders ke peeche ke genes mein mutations bhi dhoondhta hai.
PCR ke baad kitni copies?
Ek swab mein virus ke genome ki 50 copies hain, jo pehle hi DNA mein badli ja chuki hain. Maan lo har cycle DNA ko bilkul double karta hai. (a) 30 cycles ke baad kitni copies hongi? (b) Machine copies hote hi positive result ka signal deti hai. Kaun se cycle ke dauraan sample yeh threshold paar karta hai?
Pehle khud try karo. Phir solution ek-ek step karke kholo.
Do PCR results padhna
Do patients ka same PCR se test hota hai. Patient P ka sample cycle 22 par detection threshold paar karta hai; patient Q ka cycle 32 par. Kis sample mein zyada viral DNA tha, aur lagbhag kitne guna zyada? Perfect doubling maan lo.
Pehle khud try karo. Phir solution ek-ek step karke kholo.
Probe DNA ya RNA ka ek chhota single strand hai jis par ek label hota hai, pehle ek radioactive atom aur aaj aksar ek fluorescent dye. Yeh sirf us DNA se chipakta (hybridise hota) hai jiske bases iske apne bases ke complementary hon. Cloned cells ke ek set ko test karne ke liye unka DNA ek membrane par fix kiya jaata hai, probe us par se bahaya jaata hai aur jo probe bind nahi hua use dho diya jaata hai. Jahan bhi probe ne pair kiya, wahan X-ray film par (autoradiography) ya scanner mein ek spot dikhta hai. Jis clone ka gene probe wale region mein mutated hai, woh probe se pair nahi kar sakta aur koi spot nahi deta (Fig. 10.12).
3/3Autoradiography har match ke liye ek spot dikhati hai; mutant clone khaali rehta hai.
Left mein ek square membrane par do rows mein chhe gol clones hain, label hai clones on a membrane. Pehle step mein sirf yeh membrane dikhti hai. Doosre step mein laal label dot wale chhote probe strands membrane par bikhre hain, aur beech mein ek close-up dikhata hai ki ek probe DNA strand se poori tarah pair karta hai, label hai matches, binds, aur uske neeche ek probe dashed gap ke saath sirf kuch had tak pair karta hai, label hai mismatch, washes off. Aakhri step mein ek arrow right mein ek X-ray film tak jaata hai jis par clones ki positions par paanch kaale spots hain aur chhathe ki jagah ek khaali dashed circle hai, label hai no spot equals mutant.
ELISA (enzyme-linked immunosorbent assay) nucleic acids ki jagah proteins ke saath kaam karta hai. Iska key ingredient ek antibody hai jo sirf ek target ko pehchanta hai aur kisi ko nahi. Disease ke hisaab se test ya to microbe ke apne molecules, uske antigens (proteins ya glycoproteins), pakadne ke liye design hota hai, jo dikhate hain ki woh abhi maujood hai, ya un antibodies ko jo patient ne uske against banaye hain, jo dikhate hain ki immune system ka usse saamna ho chuka hai.
Ek common form, 'sandwich' ELISA, mein ek plastic well par antigen pakadne wale antibodies ki coating hoti hai. Phir enzyme se juda ek doosra antibody kisi bhi pakde gaye antigen se bind hota hai. Jab ek colourless substrate daala jaata hai, to enzyme use coloured product mein badal deta hai; rang jitna gehra, antigen utna zyada (Fig. 10.13). ELISA ka use donate kiye gaye blood ko HIV aur hepatitis B ke liye screen karne aur fever ke pehle dino mein dengue virus ka NS1 antigen pakadne mein hota hai.
4/4Enzyme colourless substrate ko coloured bana deta hai: positive result.
Do U-shaped wells aamne-saamne bane hain, labels sample aur control, har ek ke farsh par chaar Y-shaped capture antibodies khade hain. Pehle step mein caption line kehti hai antibodies coat each well. Doosre step mein diamond-shaped antigens sirf sample well mein antibodies ki arms mein baithe hain, aur line kehti hai antigen sticks if present. Teesre step mein ulte doosre antibodies, jinke upar ek gol enzyme hai, sample well ke antigens par baithe hain, aur line kehti hai enzyme-linked antibody binds. Aakhri step mein sample well ka liquid yellow ho jaata hai, label plus colour, jabki control well ka label minus clear hai, aur line kehti hai enzyme turns substrate coloured.

10.3 Transgenic Animals
Jo mouse blue light mein hara chamakta hai kyunki usmein jellyfish ka gene hai, woh ek transgenic animal (पारजीनी जंतु) hai: kisi doosre source ka gene uske DNA mein jod diya gaya hai aur wahan kaam kar raha hai. Saare transgenic animals mein 95 per cent se zyada mice hain, kyunki woh chhote hain, unhe rakhna sasta hai, woh har kuch hafton mein breed karte hain aur unke genes bahut acchi tarah samjhe gaye hain. Fish, pigs, cows, sheep, goats, rabbits aur rats ko bhi transgenic banaya gaya hai.
Aisa animal jiske DNA mein kisi doosre source ka gene jaan-boojh kar insert kiya gaya hai, aur jo us gene ka product banata hai.
ExampleHuman gene wala ek mouse jismein human disease jaise changes develop hote hain.
Ek common method mein chune gaye gene ki copies, jo ek aise promoter se judi hoti hain jo use sahi tissue mein on kare, ek fertilised egg mein inject ki jaati hain. Embryos ek foster mother mein rakhe jaate hain, aur bachchon ko test karke woh dhoondhe jaate hain jinhone gene le liya hai (Fig. 10.14). CRISPR–Cas9 jaise genome-editing tools ab aise changes zyada precisely karte hain.
4/4DNA test transgenic pups ko chun leta hai.
Pehle step mein upar left mein ek bar gene construct dikhati hai jo ek human gene se jude promoter segment se bana hai. Doosre step mein ek arrow do pronuclei wale bade fertilised egg tak jaata hai, jise left se ek blunt pipette pakde hai jabki right se ek patli needle andar jaati hai, label hai microinjection. Teesre step mein ek arrow neeche ek foster mother mouse tak jaata hai. Aakhri step mein neeche right mein teen pups dikhte hain; beech wale ka rang alag hai aur uske upar plus sign hai, label hai DNA test, one has it.
Itni mehnat kyun? Paanch reasons khaas hain (Table 10.4).
| Maksad | Kaise madad karta hai |
|---|---|
| Normal physiology aur development | Gene add karne ya badalne se pata chalta hai ki woh living body mein kya karta hai, jaise insulin-like growth factor body size ko kaise control karta hai. |
| Disease ke models | Jin animals ko disease gene diya jaata hai unmein bimari develop hoti hai, isliye treatments test kiye ja sakte hain: Alzheimer's disease, cystic fibrosis, arthritis aur kai cancers ke mouse models. |
| Biological products | Udder mein on kiya gaya human gene protein ko doodh mein daal deta hai: emphysema ke liye α-1-antitrypsin; Rosie naam ki cow ke doodh mein human α-lactalbumin (1997). |
| Vaccine safety | Jo mice human virus se infect ho sakte hain, woh har vaccine batch ko test karte hain, jaise polio vaccine testing jo monkeys ki jagah le sakti hai. |
| Chemical safety | Toxins ke liye extra-sensitive banaye gaye animals drugs, food additives aur industrial chemicals ki screening mein kam doses par aur jaldi nuksaan dikha dete hain. |
Biological-products wale raaste ko kabhi-kabhi 'pharming' kehte hain. Jo proteins itne complex hain ki bacteria unhe sahi se nahi bana sakte, unhe animal ko nuksaan pahunchaye bina uske doodh se ikattha kiya ja sakta hai, aur aise hi kaam ka lakshya phenylketonuria (PKU) aur cystic fibrosis ke treatments bhi rahe hain. Disease models bhi utne hi important hain: jo mouse human bimari develop karta hai, uski madad se researchers kisi bhi insaan par aazmaane se pehle treatment test kar sakte hain.
Left mein koi item tap karo, phir right mein uska match.

10.4 Ethical Issues
Genes ko dobara likhne ki taakat aise sawaal uthati hai jinhe akele science suljha nahi sakta. Kya research ke liye kisi animal ko bimari dena sahi hai? Kya bacterium ka gene hamare khaane mein pahunchna chahiye? Fayda kise hota hai, aur risk kaun uthata hai? Aise sawaal laboratory mein nahi, balki ethics aur law se suljhaaye jaate hain: living cheezon ke saath kya kiya ja sakta hai iske agreed standards, aur unhe laagu karne wali official bodies.
Saavdhaani ke biological reasons bhi hain. Kheton mein chhoda gaya GM organism ek ecosystem ka hissa ban jaata hai, jiske effects ka andaaza lagana mushkil hai: uske genes pollen ke zariye wild relatives tak ja sakte hain, toxin target ke alawa doosre insects ko nuksaan pahuncha sakta hai, pests mein resistance evolve ho sakta hai (jaise pink bollworm mein hua), aur khaane mein naye protein ko allergy ke liye check karna zaroori hai. Isliye har release ko case-by-case parkha jaata hai.
India mein GM research aur release Environment (Protection) Act, 1986 ke tahat 1989 mein banaye gaye rules se chalte hain. Kaam ek laboratory mein shuru hota hai jiski nigrani uski Institutional Biosafety Committee (IBSC) karti hai. Department of Biotechnology ke under Review Committee on Genetic Manipulation (RCGM) research aur chhote, confined field trials ko review karti hai. Ministry of Environment, Forest and Climate Change ke under Genetic Engineering Appraisal Committee (GEAC) bade field trials aur GM organisms aur products ko environment mein release karne ka appraisal karti hai, aur final faisla government leti hai (Fig. 10.15). Tumhari textbook committee ka purana naam use karti hai, Genetic Engineering Approval Committee; iska naam 2010 mein badla gaya.
4/4Faisla government karti hai; Bt cotton ko 2002 mein permission mili, jabki Bt brinjal abhi bhi ruka hua hai.
Chaar boxes neeche left se upar right tak seedhiyon ki tarah chadhte hain, arrows se jude hue. Pehle step mein sirf neeche wala box dikhta hai: 1 IBSC, in every lab. Doosre step mein agla box aata hai: 2 RCGM of the Department of Biotechnology, lab and confined trials. Teesre step mein teesra box aata hai: 3 GEAC of the Environment Ministry, large trials and release. Aakhri step mein sabse upar wala box aata hai: 4 Government, final decision, aur upar left mein do notes par Bt cotton allowed 2002 aur Bt brinjal on hold likha hai.
Isi system se bilkul alag nateeje nikle hain. Bt cotton 2002 mein approve hua. Bt brinjal ko 2009 mein GEAC ne clear kiya, lekin 2010 mein government ne uske release par moratorium laga diya, jo abhi tak hata nahi hai. GM mustard ke environmental release ke liye GEAC ki 2022 ki approval ko Supreme Court mein challenge kiya gaya. 2022 se, jin plants ko bina koi foreign DNA insert kiye genome-edit kiya gaya hai, unhe GMO rules se chhoot hai, aur 2025 mein India ne apni pehli do genome-edited rice varieties release ki.
Patents aur biopiracy
Sawaalon ka doosra set ownership ke baare mein hai. Patent government dwara ek limited period ke liye diya gaya right hai, India mein 20 saal, jo inventor ko doosron ko bina permission invention banane, use karne ya bechne se rokne deta hai. Patent paane ke liye invention naya ho, usmein inventive step ho aur woh useful ho. Kai deshon mein engineered microbes, genes aur processes patent kiye ja sakte hain. Mushkil tab shuru hoti hai jab koi aise plant, nuskhe ya crop variety ko patent kar le jise farmers ya tribal communities ne bahut pehle khoja aur sudhaara tha, jaise ki woh naya ho.
Basmati dikhata hai ki aisa jhagda kaise aage badhta hai. Yeh khushbudaar long-grain rice India aur Pakistan ke Indo-Gangetic plains ke farmers ki peedhiyon ki mehnat hai, ek aisa region jahan Earth par rice ki sabse zyada diversity mein se kuch hai. 1997 mein Texas ki ek company, RiceTec, ne un rice lines ka US patent le liya jo usne Basmati ko semi-dwarf rice se cross karke breed ki thin. Iske kuch claims itne broad shabdon mein the ki woh khud Basmati jaise rice tak pahunch jaate, jisse Indian farmers aur exporters chinta mein pad gaye. India ne 2000 mein patent ko challenge kiya, aur 2001 tak zyadatar claims wapas le liye gaye ya reject ho gaye, aur sirf teen specific rice lines protected rahin. Tab se India ne ek tay region ke rice ke liye Basmati ko geographical indication (GI) ke roop mein register kiya hai.
Doosre patents ne traditional Indian nuskhon ko nishana banaya. Zakhm bharne ke liye turmeric ke use par 1995 mein diya gaya ek US patent 1997 mein revoke kar diya gaya, jab India ki Council of Scientific and Industrial Research (CSIR) ne purane Sanskrit texts aur 1953 ke ek journal article se dikhaya ki yeh use bahut pehle se jaana jaata tha. Neem oil ke antifungal extract par 1994 mein diya gaya ek European patent 2000 mein revoke hua. Aise challenges aasaan banane ke liye India ne Traditional Knowledge Digital Library (TKDL) banayi, jo Ayurveda, Unani, Siddha aur yoga ke purane texts ko aise records mein badalti hai jinhe duniya mein kahin bhi patent examiners search kar sakte hain.

Naye Indian laws
Patents (Amendment) Act aur Biological Diversity Act.
Kisi desh ke plants, animals ya microbes, ya unke baare mein kisi community ke traditional knowledge, ka munafe ke liye use karna, unke maalikon ya rakhwaalon ki consent ke bina aur unhe bina payment kiye ya fayde share kiye bina.
ExampleKisi plant-based nuskhe ko patent karna jise ek community sadiyon se use karti aayi hai, uski consent ke bina aur munafa share kiye bina.
Yeh itna important kyun hai? Duniya ki biodiversity ka map dekho: India, Brazil aur Indonesia jaise tropical deshon mein zyadatar species hain, jabki zyadatar biotechnology companies aur research ka paisa North America, Europe aur Japan mein hai. Kisi vaidya ka yeh gyaan ki ek khaas patta khoon rokta hai, ek taiyaar clue hai, aur jo company use follow karti hai woh saalon ki andhi screening se bach jaati hai. Insaaf yeh maangta hai ki jis community ke gyaan se khoj shuru hui, use uski kamaai mein hissa mile (dono raaste compare karo).
Biopiracy ya fair access?
Biopiracy
- Resource ya knowledge bina permission ke liya gaya.
- Patent asli users par rok laga sakta hai.
- Koi payment ya munafe mein hissa nahi.
Fair access
- Desh aur community ki prior informed consent.
- Knowledge ke source ko credit diya jaata hai.
- Faydon ka agreed hissa: paisa, jobs ya technology.
Deshon ne laws banakar jawaab diya hai. India ne apna Patents Act 2002 mein (woh second amendment jiska zikr tumhari textbook karti hai) aur phir 2005 mein amend kiya: traditional knowledge ko invention ke roop mein patent nahi kiya ja sakta, aur applicants ko use kiye gaye kisi bhi biological material ka source aur geographical origin batana zaroori hai. Biological Diversity Act, 2002 (2023 mein amend hua) ke tahat Indian bio-resources ke foreign users, aur unke aadhaar par patent chahne wale kisi bhi insaan ko National Biodiversity Authority ki approval leni hoti hai, aur faydon ko un communities ke saath share karna hota hai jo unhe conserve karti hain. International level par Convention on Biological Diversity (1992) aur uska Nagoya Protocol (2010) access with benefit sharing ka yahi principle tay karte hain.

Summary aur key points
- Biotechnology living cells ya enzymes se bade scale par products banati hai; success catalyst, conditions aur downstream processing par depend karti hai (Fig. 10.1).
- Tissue culture totipotency ki wajah se explants se poore plants dobara ugaata hai, sucrose, salts, vitamins, amino acids, auxins aur cytokinins wale sterile medium par (Tissue culture).
- Micropropagation se hazaaron identical somaclones milte hain; meristem culture se virus-free plants (Meristems).
- Do plants ke protoplasts fuse karne se pomato jaise somatic hybrids milte hain (Hybrids).
- GM crops stress jhel sakti hain, unhe kam pesticides chahiye, harvest ke baad kam nuksaan hota hai, woh minerals ka behtar use karti hain aur zyada nutritious hoti hain (GM crops).
- Bt crops Cry protoxins banati hain jinhe insect ka alkaline gut active karta hai; toxin midgut cells mein pores banata hai. cryIAc aur cryIIAb bollworms ko target karte hain, cryIAb corn borer ko; refuges resistance ko dheema karte hain (Bt cotton).
- RNAi mein double-stranded RNA matching mRNA ko silence karta hai; Meloidogyne incognita ke against dsRNA banane wala tobacco nematode ko jhel leta hai (RNAi).
- Insulin mein chains A (21) aur B (30) hain jo disulphide bridges se judi hain; proinsulin ka C peptide hata diya jaata hai. Lilly ne chains ko E. coli mein alag-alag banakar joda (Section 10.2.1).
- Gene therapy ek working gene jodti hai: 1990 mein ADA deficiency wale ek bachche ko engineered lymphocytes diye gaye, jinhe dobara infuse karna padta tha; theek kiye gaye stem cells lambe samay tak chalte hain (Section 10.2.2).
- PCR, labelled probes aur ELISA pathogens ya mutations ko jaldi, symptoms se pehle, detect karte hain (Section 10.2.3).
- Transgenic animals, jinmein 95 % se zyada mice hain, physiology research, disease models, biological products, aur vaccine aur chemical safety testing ke kaam aate hain (Section 10.3).
- India mein GEAC GM releases ka appraisal karti hai; traditional bio-resources (Basmati, turmeric, neem) par patents ki wajah se biopiracy ke against laws bane (Section 10.4).
Practice questions answers ke saath (NCERT “Exercises”)
Ek farmer ki potato crop mein ek virus hai jo tubers ke zariye ek season se agle season tak jaata hai. Healthy stock ugaane ke liye tissue culture laboratory ko infected plant ka kaun sa hissa use karna chahiye, aur yeh kyun kaam karta hai?
Solution dikhao
AnswerApical ya axillary meristem, kyunki woh aam taur par virus se free hota hai.
Shoot-tip (apical) ya axillary meristem use karo. Viruses zyadatar vascular tissue se phailte hain, jo meristem mein abhi bana nahi hota, aur meristem cells itni tezi se divide hote hain ki virus unmein ghus nahi paata. Isliye millimetre ke chhote se hisse jitna lamba meristem, sterile medium par culture karne se, ek virus-free plant ban jaata hai jise micropropagation se multiply kiya ja sakta hai.
Gujarat ke ek grower ke paas ek shaandaar date palm hai aur woh do-ek saal mein bilkul usi jaise hazaaron plants chahta hai. Samjhao ki seed se palms ugaane ke muqable micropropagation is goal ke liye behtar kyun hai.
Solution dikhao
AnswerMicropropagation jaldi se bahut saare genetically identical plants deta hai; seeds alag-alag tarah ki santaan dete hain.
Seedlings sexual reproduction se bante hain, isliye unke genes phir se mil jaate hain aur woh us shaandaar tree se alag hote hain (date palm seedlings ke male ya female hone ki sankhya bhi pehle se nahi pata hoti). Micropropagation explants ko mitosis se multiply karta hai, isliye har plant parent ke genes wala somaclone hota hai. Yeh tez bhi hai aur ise kam jagah chahiye: ek explant ko kuch mahino mein subculture karke hazaaron plantlets banaye ja sakte hain, aur plants ko disease-free ugaaya ja sakta hai.
Tum school ke tissue-culture demonstration mein explants ugaane ke liye medium taiyaar kar rahe ho. List banao ki usmein kya-kya jaana chahiye aur har ingredient ka kaam batao.
Solution dikhao
AnswerSucrose, inorganic salts, vitamins, amino acids, auxin aur cytokinin, agar se jamaaya hua, sab sterile.
- Sucrose: carbon aur energy source, kyunki band vessel mein explant kaafi photosynthesis nahi kar paata.
- Inorganic salts: major nutrients (nitrogen, phosphorus, potassium, calcium, magnesium, sulphur) aur trace elements (iron, zinc, manganese aur doosre).
- Vitamins: metabolism ke liye coenzymes.
- Amino acids: organic nitrogen ki taiyaar supply.
- Growth regulators, ek auxin aur ek cytokinin: inka ratio tay karta hai ki callus banega, shoot ya root.
- Medium jamaane ke liye agar; medium ko use se pehle sterilise (autoclave) karna zaroori hai.
Bacillus thuringiensis apne banaye toxin se isliye bach jaata hai kyunki toxin
Answer dekho
Answer (C) crystals mein inactive protoxin bana rehta hai
Crystals mein inactive protoxin hota hai, jo sirf susceptible insect ke alkaline gut mein toxic banta hai.Human insulin ki chains banane wale E. coli cells ko transgenic kaha jaata hai. Samjhao ki ek bacterium transgenic kaise banta hai, aur short mein batao ki yeh khaas bacteria kaise banaye aur use kiye gaye.
Solution dikhao
AnswerDoosri species ka gene liye hue bacterium; jaise human insulin A- ya B-chain gene wala E. coli.
Transgenic bacterium mein kisi doosre organism se laaya gaya gene hota hai aur woh use express karta hai, aam taur par plasmid par. Example: E. coli jise human insulin ki chain A (ya chain B) ke DNA sequence wala plasmid diya gaya. Bioreactor mein ugaane par har culture ek chain banata hai; chains ko purify karke disulphide bridges se joda jaata hai, jisse natural hormone jaisa identical human insulin milta hai (Section 10.2.1 dekho).
Tumhari gaon ki panchayat bahas kar rahi hai ki farmers ko GM crop ugaani chahiye ya nahi. Fayde aur nuksaan ki ek balanced list banao, aur har nuksaan ke liye ek safeguard suggest karo.
Solution dikhao
Fayde: chemical pesticides ka kam spray (jaise Bt cotton), drought, salt ya thand jhelne ki kshamta, harvest ke baad kam nuksaan, fertiliser ka behtar use, zyada nutritious food (golden rice), aur industrial raw materials dene wali crops.
Nuksaan aur safeguards:
- Pests mein resistance evolve ho sakta hai (pink bollworm): refuges lagao aur do ya zyada toxins wali crops use karo.
- Genes pollen se wild relatives tak phail sakte hain: isolation distances rakho aur khet ke kinaron ko monitor karo.
- Non-target insects ko nuksaan ho sakta hai: approval se pehle effects test karo aur release ke baad monitor karo.
- Naya protein allergy kar sakta hai: approval se pehle food ko allergens ke liye test karo.
- Seed mehenga hai aur har season khareedna padta hai (hybrids): public-sector varieties aur price regulation.
- Patents se control kuch companies ke haath mein simat sakta hai: farmers' rights aur benefit sharing par laws.
Ek accha answer dono lists ko taulta hai aur batata hai ki India mein har GM crop ko release se pehle RCGM aur GEAC ka review paar karna padta hai.
Ek cotton farmer poochhti hai ki bollworms uske Bt cotton ko kyun chhod dete hain. Use samjhao ki Cry protein kya hai, yeh originally kis organism se aata hai, yeh bollworm ko kaise maarta hai, aur yeh uske plants ke andar kaise pahuncha.
Solution dikhao
AnswerBacillus thuringiensis; alkaline gut mein active hokar yeh midgut cells mein pores banata hai; sprays aur Bt crops mein use hota hai.
Cry proteins Bacillus thuringiensis ke banaye insecticidal crystal proteins hain. Bacterium mein yeh inactive protoxin crystals ke roop mein store hote hain. Bollworm ke alkaline midgut mein crystals ghul jaate hain aur gut enzymes active toxin release karte hain, jo midgut epithelial cells ke receptors se bind hokar pores banata hai, isliye cells phool kar phat jaate hain aur larva mar jaata hai. Logon ne pehle Bt ko biological spray ki tarah use kiya; ab cryIAc aur cryIIAb jaise cry genes cotton mein daale jaate hain (aur cryIAb maize mein), isliye plant apni protection khud karta hai aur bahut kam insecticide chahiye.
Ek bachchi ke blood mein lagbhag bilkul adenosine deaminase nahi hai. Step-by-step batao ki 1990 ki gene therapy team ne aise patient ka treatment kaise kiya, aur isse samjhao ki gene therapy kya hai. Woh treatment permanent cure kyun nahi tha, aur lambe samay ka cure kaise paaya ja sakta hai?
Solution dikhao
Gene therapy patient ke cells mein gene ki working copy pahunchakar genetic defect ko theek karti hai, taaki woh faulty copy ka kaam sambhal le.
- ADA deficiency: enzyme adenosine deaminase missing hota hai, toxic by-products lymphocytes ko maar dete hain aur immune system fail ho jaata hai (SCID ka ek form).
- Patient ke blood se lymphocytes lekar culture mein ugaaye gaye.
- Ek retroviral vector functional ADA cDNA ko in lymphocytes mein le gaya.
- Engineered cells patient ko wapas diye gaye aur unhone ADA banaya.
Lymphocytes amar nahi hote, isliye infusions dohrane padte the. Lambe samay ke cure ke liye gene bone marrow ke blood banane wale stem cells mein hona chahiye, jo naye lymphocytes banate rehte hain; aisa treatment 2016 mein Europe mein approve hua. (Early embryo ke cells ko theek karna bhi permanent hoga, lekin isse germline badal jaayegi aur India mein iski permission nahi hai.)
E. coli mein growth hormone jaise kisi human gene ko clone aur express karne ke steps ka flow diagram banao. Shuruaat in steps ko sahi order mein lagakar karo.
- Cells ko bioreactor mein ugaao
- Plasmid ko E. coli mein daalo
- cDNA aur plasmid ko ek hi enzyme se kaato
- Unhe DNA ligase se jodo
- Hormone ko purify karo
- Antibiotic par transformed cells select karo
- Hormone ke mRNA se cDNA banao
Solution dikhao
Tumhare diagram mein arrows se jude saat boxes hone chahiye: pituitary mRNA → reverse transcriptase se cDNA → cDNA aur plasmid same restriction enzyme se kaate gaye → DNA ligase se jode gaye (recombinant plasmid, gene ke aage bacterial promoter ke saath) → competent E. coli ka transformation → antibiotic medium par selection → bioreactor mein bade scale par culture → growth hormone purify karne ke liye downstream processing.
Seeds mein oil mainly triglycerides (teen fatty acids se juda glycerol) ke roop mein store hota hai. Recombinant DNA technology ka use karke suggest karo ki tum bahut kam oil wala seed kaise bana sakte ho, aur batao ki seed uski jagah kya store kar sakta hai.
Solution dikhao
AnswerTriglyceride synthesis ke liye zaroori seed genes ko silence ya knock out karo; tab carbon starch, sugars ya protein mein jaata hai.
Triglycerides enzymes ki ek chain se bante hain, jiske end mein diacylglycerol acyltransferase (DGAT) hai, jo teesra fatty acid jodta hai. Koi aisa construct insert kiya ja sakta hai jo DGAT gene ke against, ya WRINKLED1 jaise seed oil synthesis ke master regulator ke against, dsRNA (RNAi) ya antisense RNA banaye, aur use seed-specific promoter ke peeche rakha jaaye taaki sirf seeds par asar ho. Iski jagah genome editing se gene ko knock out bhi kiya ja sakta hai. Oil synthesis block hone par seed apna carbon starch, sugars ya protein mein laga dega. Aise seeds shayad kam taakat se germinate karein, kyunki oil unka energy store hai.
GM crops par bahas mein golden rice ka zikr aksar hota hai. Online ya library mein ispar research karo: yeh kya banata hai, kaun se genes jode gaye, ise kyun develop kiya gaya, aur aaj yeh kahan khada hai? Apne findings ko solution se check karo.
Solution dikhao
- Yeh kya banata hai: endosperm mein, jo hissa hum khaate hain, β-carotene (provitamin A), jisse daanon ko golden rang milta hai.
- Jode gaye genes: ek phytoene synthase gene (psy, pehle version mein daffodil se aur Golden Rice 2 mein maize se) aur ek soil bacterium ka carotene desaturase gene (crtI).
- Kyun: vitamin A deficiency se night blindness hoti hai aur immunity kamzor hoti hai, jisse rice khaane wale deshon mein kai chhote bachche mar jaate hain ya andhe ho jaate hain.
- Status: Philippines ne ise 2021 mein kheti ke liye approve kiya, lekin wahan 2024 ke ek court order ne commercial planting rok di; India mein yeh nahi ugaaya jaata.
Kya human blood mein aise enzymes hote hain jo proteins aur nucleic acids ko todte hain? Tumhare answer ka us doctor ke liye kya matlab hai jo nas mein protein medicine ya DNA ka tukda inject karna chahta hai?
Solution dikhao
AnswerHaan. Inject kiye gaye proteins aur naked DNA dheere-dheere toot jaate hain, isliye unhe protection ya baar-baar doses chahiye.
Haan. Plasma mein proteases hote hain, jaise clotting aur clot-gholne wale systems ke thrombin aur plasmin (jo zyadatar inactive precursors ke roop mein ghoomte hain), aur DNase aur RNases jaise nucleases. Isliye inject ki gayi protein drug ghanton se dino mein toot kar hat jaati hai, aur naked DNA ya RNA aur bhi jaldi nasht ho jaata hai. Isiliye protein drugs baar-baar doses mein di jaati hain ya zyada der tak chalne ke liye chemically modify ki jaati hain, aur genes ko viral vectors ya lipid particles ke andar pahunchaya jaata hai.
Researchers aisa insulin banane ki koshish kar rahe hain jise nigla ja sake. Aazmaaye ja rahe ek approach ka pata karo aur woh main rukawat batao jo kisi bhi oral protein medicine ko paar karni padti hai.
Solution dikhao
Rukawatein: stomach acid aur protein pachane wale enzymes pepsin, trypsin aur chymotrypsin protein ko amino acids mein tod dete hain, aur poora protein bhi itna bada hota hai ki gut lining aasaani se paar nahi kar paata, isliye blood tak bahut kam pahunchta hai.
Aazmaaye ja rahe approaches: enteric-coated capsules jo sirf intestine mein ghulte hain, drug ke saath pack kiye protease inhibitors, absorption enhancers, nanoparticles aur aise modified insulins jo digestion jhel lein. Isse juda ek success hai oral semaglutide (2019 mein approve), type 2 diabetes ki ek peptide medicine jo absorption enhancer ke saath li jaati hai. Jab yeh chapter likha gaya tab koi oral insulin routine use mein nahi tha.

Answer (D) Micropropagation – protoplasts fuse karna
Protoplasts fuse karna somatic hybridisation hai; micropropagation explants ko multiply karke identical plants banata hai.E. coli mein bane human insulin ko pig insulin se zyada isliye pasand kiya jaata hai kyunki yeh
Answer dekho
Answer (A) bilkul human amino acid sequence wala hai
Identical sequence ka matlab hai kam allergic aur immune reactions; mature insulin, chahe kisi bhi source se ho, mein disulphide bridges hote hain aur C peptide nahi hota.
